Work on a similar project across the Baltic Sea in Finland started in 2015. The project is expected to cost about 19 billion Swedish kroner (1.5 billion pounds $1.8 billion) and create 1,500 jobs, although construction will take decades. The humid conditions in Oskarshamn are perfect for testing nuclear waste containers, says Ylva StenqvistĮarlier this year, the Swedish government approved plans for a true geological disposal facility (GDF) to be built in Forsmark, some 150km north of Stockholm. “So we deliberately chose this location to speed up some of the experiments, really put our materials and our methods under stress and see how they hold up in this pretty aggressive environment.” “Because if we conduct our experiments in a really dry area, we have to wait forever for any results. “This side was chosen because it’s quite wet,” she explains. But according to Ylva Stenqvist, project manager at the country’s nuclear power plant operator, SKB, it’s perfect for testing. Such a humid environment would not be suitable for a real repository. The bedrock here is rugged and flows through salt water – ancient brine that has migrated far up from the Baltic Sea over thousands of years. It is used for experiments studying how waste can be packaged and buried, and how the materials used might degrade over time. Hundreds of meters below the surface, a network of huge man-made caves has been dug into the rock. Much of the research was carried out at the Aspo Hard Rock Laboratory, a facility built near Oskarshamn in the south of the country. Sweden has experimented with storing nuclear waste underground at the Aspo Hard Rock Laboratory This is a process known as geological disposal, and the country’s scientists have spent decades studying various ways it might be carried out. It plans to bury its waste deep underground in rocks and leave it there forever. Sweden has already drawn its own conclusions. “So the surface of the earth and human civilizations are changing much faster than the rate at which the radioactivity in this spent nuclear fuel can decay.” ![]() The last ice age ended about 10,000 years ago. “The Roman Empire lasted about 500 years. “We cannot rely on institutional control for periods much longer than a few centuries,” says Prof. ![]() “After about 1,000 years, about 10% of the original radioactivity remains, which will slowly decay over about 100,000 years.” ‘Spent fuel is highly radioactive and this radioactivity takes a long time to decay,’ explains Prof Neil Hyatt, senior scientific adviser to the UK’s Nuclear Waste Services. The question of what to do with it afterwards has occupied many governments, including the British one, for years.Įven after around 60 years of commercial and military programmes, the UK stockpile of the most hazardous high-level waste is a few thousand tonnes, although there will also be several hundred thousand tonnes of intermediate-level waste that will also need to be treated. Nuclear waste must be cooled before it can be shipped to other storage sites Intense radioactivity generates a lot of heat, and this type of material needs to be chilled for a long time before it can be removed for storage.

Aβ results from the proteolytic processing of the amyloid precursor protein (APP), a transmembrane protein expressed in neuronal and non-neuronal cells in the CNS. Extracellular senile plaques are formed from Aβ peptides of different lengths, as follows: the 40-residue peptide Aβ 40 represents the most abundant physiological Aβ isoform in the brain, while the pathogenic 42-residue Aβ 42 is elevated in AD brains ( Näslund et al., 1994). ![]() Pharmacological strategies targeting the proteolytic cleavage of APP disrupt the relationship between extracellular and intracellular Aβ, providing a therapeutic approach for the disease.Īlzheimer's disease (AD) is a chronic neurodegenerative disorder characterized by a progressive cognitive impairment and by the presence of the following two characteristic lesions of the cerebral cortex: neurofibrillary tangles of tau and extracellular deposits of β-amyloid (Aβ) peptides as senile plaques ( Cipriani et al., 2011). The other pool is released into the extracellular space and contributes to the propagation of the pathology from diseased to healthy neurons. One pool accumulates inside the cytosol, inducing the loss of synaptic plasticity potential. Our results also suggest that subsequent to APP cleavage two pools of Aβos are produced. We showed that extracellular Aβos propagate excitatory synaptic alterations by promoting amyloid precursor protein (APP) processing. SIGNIFICANCE STATEMENT Here we provide the proof that a vicious cycle between extracellular and intracellular pools of Aβ oligomers (Aβos) is required for the spreading of Alzheimer's disease (AD) pathology. Together, our data unveil a complementary role for Aβos in AD, while intracellular Aβos alter synaptic function, extracellular Aβos promote a vicious cycle that propagates synaptic pathology from diseased to healthy neurons. Furthermore, we provide evidence that following APP processing, extracellular release of Aβos mediates the propagation of the synaptic pathology characterized by a decreased spine density of neighboring healthy neurons in an APP-dependent manner. Intracellular Aβos participate in synaptic dysfunctions as shown by pharmacological inhibition of APP processing or by intraneuronal infusion of an antibody raised against Aβos. We demonstrated that Aβos/APP interaction increases the amyloidogenic processing of APP leading to intracellular accumulation of newly produced Aβos. These alterations were not observed in APP KO neurons, suggesting that APP expression is required. We showed that extracellular application of Aβos reduced glutamatergic synaptic transmission and long-term potentiation. We used complementary techniques ranging from electrophysiological recordings and molecular biology to confocal microscopy in primary cortical cultures, and from acute hippocampal and cortical slices from male wild-type and amyloid precursor protein (APP) knock-out (KO) mice to assess the effects of Aβos on glutamatergic transmission, synaptic plasticity, and dendritic spine structure. Here, we investigated the mechanism by which Aβos affect the function of synapses and how these alterations propagate to surrounding healthy neurons. This pathologic feature is driven by amyloid-β oligomers (Aβos) and propagates from neuron to neuron. Alterations of excitatory synaptic function are the strongest correlate to the pathologic disturbance of cognitive ability observed in the early stages of Alzheimer's disease (AD). ![]() Add in the D-BOX’s crystal-clear analog summing, and even the smallest project studios can have the best monitoring and mixing technology available in just one elegant and easy to use rack unit. In 1965 Évry became part of the French new town initiative and took its current name. Évry had just been chosen to become a " new town" of the suburbs of Paris, destined to host tens of thousands of suburbanites, and so the name "Petit-Bourg" (literally meaning "little borough, small town" in modern French, although etymologists think that this name was in fact the corruption of an old Gallic word with a totally different meaning) was deemed too old fashioned and improper for the new large suburban city of Évry to be built. On 29 June 1965 the name of the commune was shortened into "Évry" only. The factory owed its name to the hamlet of Petit-Bourg (one of the three hamlets on the territory of Évry) where it was built. In 1881 the name of the commune was changed into Évry-Petit-Bourg at the request of entrepreneur Paul Decauville, owner of Ateliers de Petit-Bourg, a large boiler works located in Évry and at the time the largest employer in the area. After the conquest, the name was corrupted into Latin Apriacum, then Medieval Latin Avriacum, and later Evriacum.
